Standard RIPA clears the cytosol and most membrane-associated protein. Detergent-resistant membrane (DRM) and lipid raft material stays in the pellet. You can force it out with 2% SDS — but the protein comes out denatured.

Neither buffer ships with protease inhibitors. Add your cocktail — and phosphatase inhibitors if you are reading phosphorylation state — immediately before use.
RIPA Buffer contains SDS and deoxycholate. Confirm detergent tolerance for your protein quantitation method and for any antibody-based step that needs native conformation.
Both products arrive as 1x working solutions and are used as received. There is nothing to reconstitute, so there is no preparation step to vary between users.
Both buffers are stored at 4°C and carry a 1 year shelf life from the date of receipt. Neither is intended for clinical, diagnostic, or therapeutic use.
