What each buffer recovers

Solubilization reach, side by side

Standard RIPA clears the cytosol and most membrane-associated protein. Detergent-resistant membrane (DRM) and lipid raft material stays in the pellet. You can force it out with 2% SDS — but the protein comes out denatured.

If your target is soluble or membrane-associated

RIPA Buffer

The everyday choice for whole-cell and tissue lysis feeding Western blot, immunoprecipitation, and ELISA sample prep. Fully disclosed formulation, no protease inhibitors added, so you pick the cocktail.

If your target sits in the RIPA-insoluble pellet

UltraRIPA™ Buffer

Recovers more than 70% of protein from that pellet without denaturing it — so lipid raft and other DRM-associated proteins stay usable for immunoprecipitation and enzyme activity assays.

Running both is common: lyse with RIPA Buffer, spin, then re-extract the pellet with UltraRIPA™ Buffer to get the fraction the first pass left behind.

Working notes

Add inhibitors yourself

Neither buffer ships with protease inhibitors. Add your cocktail — and phosphatase inhibitors if you are reading phosphorylation state — immediately before use.

Check assay compatibility

RIPA Buffer contains SDS and deoxycholate. Confirm detergent tolerance for your protein quantitation method and for any antibody-based step that needs native conformation.

No dilution step

Both products arrive as 1x working solutions and are used as received. There is nothing to reconstitute, so there is no preparation step to vary between users.

Store at 4°C on arrival

Both buffers are stored at 4°C and carry a 1 year shelf life from the date of receipt. Neither is intended for clinical, diagnostic, or therapeutic use.

Yes. A sequential extraction is the standard way to separate the two fractions: lyse in RIPA Buffer, clear by centrifugation, then re-extract the remaining pellet with UltraRIPA™ Buffer to recover the DRM / lipid raft material.
No. Both are supplied without protease inhibitors so you can match the cocktail to your target proteins. Add inhibitors immediately before use.
No. UltraRIPA™ Buffer is a proprietary detergent formulation and its pH, molarity, and component breakdown are not published. RIPA Buffer, by contrast, is fully specified: 50 mM Tris-HCl at pH 8.0, 150 mM NaCl, 1% NP-40 alternative, 0.1% SDS, and 0.5% sodium deoxycholate.
