A buffer widely used for cell lysis and protein extraction.
Whereas sodium dodecyl sulphate (SDS)-containing buffer enable us to extract proteins immensely from tissues or cells, it disrupts protein structures. Although buffer containing 1% Triton X-100 keeps the protein structure and function, the extraction efficiency for membrane proteins is not sufficient. RIPA (Radio-ImmunoPrecipitation Assay) buffer have so high protein extraction ability and low protein denaturation that it can be applied to various applications. Therefore, RIPA buffer is one of the most useful buffer for protein extraction from tissues and cells.
Formulation: 50 mM Tris-HCl (pH8.0), 150 mM NaCl, 1% NP-40 alterative, 0.1% SDS and 0.5% sodium deoxycholate)
