Description:
PBST with 5% Nonfat Dry Milk is a commonly used blocking and antibody dilution solution for Western blotting. Milk contains many proteins of various sizes, giving a better chance to reduce background banding compared to single-protein solutions like BSA. It works well with a wide range of antibodies, including many phospho-specific and total antibodies. PBS helps maintain a stable pH, which is important for optimal antibody-antigen interactions and overall assay performance. The phosphate in PBS may interfere with the detection of phosphorylated proteins. Tween-20 in PBST helps reduce background and non-specific binding.
* The optimal concentration and blocking time may vary depending on the specific experimental conditions and antibodies used.
Usage consideration:
- Not recommended for detecting phosphorylated proteins due to potential interference from phosphate in PBS.
- May interfere with biotin-based detection systems due to endogenous biotin in milk.
Applications
- Block non-specific binding: The nonfat dry milk proteins occupy vacant binding sites on the membrane, reducing background noise and improving the signal-to-noise ratio in Western blots.
- Dilute antibodies: It serves as a medium for diluting primary and secondary antibodies used in immunoblotting procedures.

Quality Control:
- Appearance: Clear, Colorless Liquid.
- pH: 7.4
- Sterility: Filtered 0.1 micron Twice and 0.04 micron once in a sterile environment.
- DNase activity: None was detected after incubation of plasmid DNA and this product for 18 hr at room temperature.
- RNase activity: No RNase activity was detected after incubation of ribosomal RNA and this product for 18 hr at room temperature.
