Phosphate-Buffered Saline (PBS) with 0.05% Tween-20 (PBST) is a widely used buffer in ELISA and immunostaining procedures due to its effective properties in reducing non-specific binding and enhancing washing efficiency. Tween 20 acts as a non-ionic detergent-based blocking agent, preventing the non-specific binding of antibodies or proteins to surfaces like microplate wells or membranes. This effect reduces background noise and increases the signal of interest. This reduces background noise and improves signal detection in ELISA and Western blotting assays. It is a non-ionic detergent that helps to disrupt non-specific protein-protein interactions during washing steps, enhancing the removal of unbound proteins or antibodies. It also reduces surface tension, allowing for better penetration and removal of unbound molecules.
Applications:
- Washing Buffer: PBST is commonly used as a washing buffer in ELISA assays. The addition of Tween-20, a non-ionic detergent, helps to remove non-specifically bound proteins from the microplate wells, thereby reducing background noise and improving the specificity of the assay results.
- PBST is used in conjunction with other blocking agents like BSA to block unsaturated binding sites on the microplate surface, further minimizing non-specific interactions.
- In immunohistochemistry and other immunostaining techniques, PBST serves as an effective washing solution that helps to remove unbound antibodies, thus preventing non-specific staining and enhancing signal clarity.

All DiagnoCine Precision Sterile buffers are filtered-sterilized with a 0.1-micron filtration two times and 0.04-micron filtration one time. Thus, mycoplasma contamination is prevented. The smallest size mycoplasma type can be about 0.2 microns.
