Description:

Malt Extract Agar (MEA) plates with streptomycin sulfate are selective fungal isolation plates in which a standard MEA formulation is supplemented with an aminoglycoside antibiotic (streptomycin) to inhibit bacteria while allowing yeasts and molds to grow on a carbohydrate-rich, mildly acidic medium.  MEA uses malt-derived carbohydrates (maltose, dextrins, sometimes added glucose or glycerol) plus vitamins and other growth factors as the primary carbon and energy sources, strongly supporting fungal metabolism and sporulation.  Peptone or mycological peptone provides amino acids and nitrogenous compounds promoting luxuriant fungal growth with typical colony morphology and pigmentation.

Streptomycin is an aminoglycoside antibiotic that binds to the 30S subunit of the bacterial ribosome, causing misreading of mRNA and blocking accurate protein synthesis.  Streptomycin shows strong activity against many gram-negative and some gram-positive bacteria with minimal effect on fungi.  The acidic pH and fungal nutrients (malt extract plus peptone) bias growth toward fungi and away from many bacteria.  Streptomycin sulfate further suppresses bacterial flora that could still grow under these conditions, reducing bacterial background and preventing confluent bacterial films that would otherwise obscure fungal colonies.
