Description:
Malt Extract Agar (MEA) plates with ampicillin are selective fungal culture plates where MEA is supplemented with β-lactam antibiotic (ampicillin) to inhibit bacteria while permitting yeasts and molds to grow on the carbohydrate-rich, mildly acidic medium. MEA contains malt extract as the primary carbon and energy source, supplying maltose, dextrins, vitamins, and growth factors that strongly support fungal metabolism and sporulation. Mycological peptone provides amino acids and other nitrogenous compounds that promote luxuriant fungal growth with typical morphology and pigmentation.
Ampicillin is a β-lactam antibiotic that binds to penicillin-binding proteins (PBPs, including transpeptidases) in the bacterial cell wall, blocking peptidoglycan cross-linking, weakening the wall, and leading to osmotic lysis and bacterial death. Ampicillin has a good activity against many gram-positive and some gram negative bacteria, targeting cell wall synthesis. The acidic pH and fungal nutrients (malt extract, peptone) favors growth toward yeasts and molds, and ampicillin further suppresses bacterial flora that may tolerate lower pH, reducing bacterial background and overgrowth on the plate surface.
