Columbia Agar Base with carbenicillin is a nutrient-rich, non-blood agar medium in which standard Columbia agar base is supplemented with the β-lactam antibiotic carbenicillin to introduce selectivity through inhibiting carbenicillin-susceptible bacteria while permitting growth of carbenicillin-resistant organisms. Columbia agar base is a complex peptone medium originally devised as a blood-agar base for the cultivation of fastidious organisms such as streptococci, staphylococci, Neisseria, and Haemophilus, as well as non-fastidious bacteria. The peptones and yeast extract provide amino acids, peptides, nitrogen, B-complex vitamins, and other growth factors that support bacterial growth and well-defined colony morphology. Starch works as a carbon and energy source and also is able to help neutralize toxic metabolites, aiding the recovery of more delicate organisms, while NaCl maintians the osmotic balance.
Carbenicillin is a broad-spectrum β-lactam (a carboxypenicillin) that inhibits bacterial cell wall synthesis by binding penicillin-binding proteins and blocking peptidoglycan cross-linking, leading to lysis of actively growing susceptible cells. When carbenicillin is added to Columbia Agar Base, the medium becomes selective by inhibiting many carbenicillin-susceptible Gram-negative rods and Gram-positive bacteria, while organisms that are intrinsically resistant or carry β-lactamase or other carbenicillin-resistance determinants can still grow.
