Description:
Columbia agar base is a complex peptone medium originally devised as a blood-agar base for fastidious organisms such as streptococci, staphylococci, Neisseria, and Haemophilus as well as non-fastidious bacteria. It typically contains a blend of special peptones from casein and meat (often with heart extract), yeast extract, starch (corn/potato), sodium chloride, and agar at about neutral pH. The peptones and yeast extract provide amino acids, peptides, nitrogen, vitamins (including B-complex), and other growth factors that support rapid, luxuriant bacterial growth and good colony morphology. Starch acts as an energy source and can bind or neutralize toxic metabolites, helping the recovery of more delicate organisms, while sodium chloride maintains osmotic balance and agar supplies the solid surface for colony development.
Kanamycin is an aminoglycoside antibiotic that binds to the 30S ribosomal subunit and interferes with protein synthesis, causing misreading of mRNA and eventual death of susceptible bacteria. When kanamycin is added to Columbia Agar Base, the medium becomes selective: many kanamycin-susceptible Gram-negative rods and some Gram-positive bacteria are inhibited, whereas strains that are intrinsically resistant or carry kanamycin-resistance determinants (such as aminoglycoside-modifying enzymes or rRNA/ ribosomal protein mutations) can still grow.
Columbia Agar Base with kanamycin is used when you want the strong growth support and colony morphology of Columbia agar but need to suppress kanamycin-susceptible background flora, enriching for kanamycin-resistant organisms. Typical uses include maintaining or isolating kanamycin-resistant laboratory strains, selecting resistant mutants, or recovering target organisms from mixed clinical, environmental, food, or water samples where competing susceptible bacteria would otherwise overgrow.
