Description:
Malt Extract Agar (MEA) plates with Penicillin G are selective fungal plates in which the standard MEA formulation is supplemented with β-lactam antibiotic (penicillin G) to inhibit bacteria while allowing yeasts and molds to grow on a nutrient-rich, mildly acidic surface. MEA uses malt extract as the primary carbon and energy source, typically around 20–30 g/L, providing maltose, glucose, dextrins, vitamins, and growth factors that strongly support fungal metabolism and sporulation. A small amount of peptone or mycological peptone (often 1–5 g/L in many MEA formulations) supplies amino acids and other nitrogenous compounds, giving luxuriant fungal growth with typical colony morphology and pigmentation.
Penicillin G is a β-lactam antibiotic that binds to bacterial penicillin-binding proteins (PBPs), which include transpeptidases responsible for cross-linking peptidoglycan strands in the cell wall. By inactivating PBPs, Penicillin G blocks peptidoglycan cross-linking, weakens the bacterial cell wall, and leads to osmotic lysis and bacterial death, especially in actively growing gram-positive organisms. Fungal cells lack peptidoglycan cell walls, so at typical microbiological plate concentrations penicillin G selectively targets bacteria and leaves yeasts and molds largely unaffected.
The acidic pH and fungal nutrients (malt extract plus peptone) already biases growth toward yeasts and molds, reducing the competition of many bacteria relative to fungi. The addition of Penicillin G to MEA further suppresses susceptible gram-positive bacteria and some gram-negative bacteria.
