Description
Tryptic Soy Agar (TSA) is a versatile, nutrient-rich medium widely used for the cultivation, isolation, and enumeration of a broad range of microorganisms, including both fastidious and non-fastidious species. Its balanced formulation-featuring tryptone and soy peptone-delivers essential amino acids, peptides, vitamins, and minerals, while glucose provides an easily metabolized carbohydrate source. Phosphate ensures optimal pH stability, and sodium chloride maintains osmotic equilibrium.
The addition of streptomycin sulfate at 50 µg/mL transforms TSA into a selective medium, effectively inhibiting the growth of streptomycin-sensitive bacteria. This selective property is especially valuable for isolating and propagating streptomycin-resistant strains, screening for resistance, or suppressing background flora in mixed cultures. Such selectivity is particularly useful in clinical, food, water, and environmental microbiology, as well as in research applications involving antibiotic susceptibility testing and genetic studies.
TSA can also be further enriched with blood or other additives to support the growth of more fastidious organisms. Its reliability and flexibility make it a staple in microbiological laboratories for routine cultivation, maintenance, and quality control of microbial cultures.
