Potato Dextrose Agar (PDA) plates with streptomycin sulfate are a selective fungal medium formulated to support robust growth of yeasts and molds while inhibiting many co-present bacteria by adding an aminoglycoside antibiotic to a fungally favorable base. PDA contains potato infusion/extract, dextrose, and agar, yielding a carbohydrate-rich, slightly acidic medium that promotes luxuriant mycelial growth, sporulation, and pigment production in a broad range of environmental, food, and cosmetic yeasts and molds. The potato infusion provides complex nutrients and growth factors, dextrose supplies an easily metabolized carbon source, and agar forms a firm, clear gel suitable for colony isolation and morphology assessment.
Streptomycin is an aminoglycoside antibiotic that binds to the 30S subunit of bacterial ribosomes, disrupting protein synthesis and leading to growth inhibition or death of susceptible bacteria, particularly many gram-negative rods and some gram-positive species. When incorporated into PDA at typical working concentrations (commonly around 100 µg/mL = 100 mg/L), streptomycin markedly reduces bacterial colony formation while leaving most filamentous fungi and yeasts largely unaffected, since fungi lack the bacterial-type ribosomes that streptomycin targets. PDA already favors fungi over many bacteria due to its nutrient profile and mildly acidic pH; the addition of streptomycin further suppresses susceptible bacteria, creating a selective environment where fungal colonies can grow, spread, and sporulate without being overgrown by bacterial contaminants.
