Columbia Agar base is a highly nutritious, general-purpose medium developed for the isolation and cultivation of fastidious and non-fastidious microorganisms from a variety of clinical and non-clinical materials. It is frequently used as a base for selective media formulations, where different combinations of antimicrobial agents can be added to enhance selectivity for specific organisms.
The rich nutrient composition—derived from a blend of peptones, heart and meat extracts, and yeast extract—provides essential amino acids, vitamins (including those of the B complex), and other growth factors necessary for luxuriant bacterial growth. Sodium chloride maintains osmotic balance, while maize starch serves as an energy source and helps neutralize toxic metabolites.
When supplemented with Penicillin G, a β-lactam antibiotic, Columbia Agar base becomes more selective by inhibiting many penicillin-sensitive Gram-positive bacteria and some susceptible Gram-negative species, depending on their intrinsic and acquired resistance.. This allows for improved isolation and detection of penicillin-resistant bacteria and certain gram-negative species in samples with mixed microbial populations. The medium can be applied to samples such as food, dairy, water, and pharmaceuticals, especially in contexts where penicillin-susceptible background flora need to be suppressed
Columbia Agar base with Penicillin G is especially useful in applications where suppression of background flora is needed to recover target organisms, such as in the detection of resistant strains or the isolation of specific pathogens from complex specimens.
