Description:


Potato Dextrose Agar (PDA) plates with penicillin G are a selective fungal medium designed to promote vigorous growth of yeasts and molds while inhibiting many co-present bacteria via a beta-lactam antibiotic.  PDA contains potato infusion/extract, dextrose, and agar, giving a carbohydrate-rich, slightly acidic environment (around pH 5.4–5.8) that supports luxuriant mycelial growth, sporulation, and pigment production in a wide range of environmental, food, and cosmetic yeasts and molds.  The potato component supplies complex nutrients and growth factors; dextrose provides an easily metabolized carbon source; agar forms a firm gel that allows clear colony isolation and morphology assessment


Penicillin G is a beta-lactam antibiotic that targets bacterial cell wall synthesis by binding penicillin-binding proteins, leading to lysis of susceptible bacteria, especially many gram-positive and some gram-negative species.  When incorporated into PDA at an appropriate concentration (commonly in the tens of µg/mL range as part of a penicillin-streptomycin–type cocktail), penicillin G suppresses growth of susceptible bacteria, reducing bacterial background without significantly affecting most filamentous fungi and yeasts, which lack peptidoglycan cell walls.  
PDA’s mildly acidic pH and nutrient profile already favor fungal growth over many bacteria; adding penicillin G further decreases bacterial colony formation, making fungal colonies easier to detect, enumerate, and subculture from mixed samples.
