Description:

Columbia Agar Base with streptomycin sulfate is a nutritionally rich, non-blood agar medium in which standard Columbia agar base is supplemented with the aminoglycoside antibiotic streptomycin to make the medium selective by suppressing susceptible bacteria while allowing streptomycin-resistant organisms to grow.  Columbia agar base is a complex peptone medium originally devised as a blood-agar base for the cultivation of fastidious organisms (e.g., streptococci, staphylococci, Neisseria, Haemophilus) as well as non-fastidious bacteria. It typically contains special peptones, yeast extract, maize/corn starch, sodium chloride, and agar at a near-neutral pH. The peptones and yeast extract provide amino acids, peptides, nitrogen, vitamins (including B-complex), and other growth factors that support luxuriant bacterial growth and good colony morphology. Maize starch serves as an energy source and helps neutralize toxic metabolites, aiding the recovery of more delicate organisms, while sodium chloride maintains osmotic balance and agar provides the solid support for colony development.

Streptomycin is an aminoglycoside antibiotic that binds to the 30S ribosomal subunit and interferes with protein synthesis, causing misreading of mRNA and ultimately bacterial cell death in susceptible organisms. When streptomycin sulfate is added to Columbia Agar Base, the medium becomes selective: many streptomycin-susceptible Gram-negative and some Gram-positive bacteria are inhibited, whereas strains that are intrinsically resistant or that carry streptomycin-resistance determinants can still grow.
