Description:
Hektoen Enteric Agar (HE) with tetracycline is a modified moderately selective and differential plate medium in which the standard HE formulation is supplemented with a broad-spectrum protein synthesis inhibitor that further suppresses tetracyline-susceptible background flora. HE contains bile salts and dyes that selectively inhibit most Gram-positive organisms. Most non-pathogenic coliform flora are partially or fully inhibited, and this allows enteric pathogens that are able to tolerate the bile and dyes to grow more readily. HE also contains carbohydrates, and fermenting non-target enterics produce acid shifting colony and medium color to yellow to orange/salmon around the colonies. With carbohydrate fermentation salmonella and shigella act as non-fermenters relying on peptone and as a result, they produce alkaline products that make the colony and surrounding medium green to blue-green. Sodium thiosulfate and ferric ammonium citrate help detect H2S by forming black ferrous sulfide, Many Salmonella will have black centers in their colonies.
Tetracycline is a broad-spectrum bacteriostatic antibiotic that binds to the 30S ribosomal subunit and blocks aminoacyl-tRNA binding, inhibiting protein synthesis in susceptible bacteria. It inhibits many Gram-positive and Gram-negative organisms, including numerous enteric commensals. The combination of HE and tetracycline allow for the further suppression of tetracycline-susceptible Gram-negative rods and Gram-positives that might otherwise grow despite bile and dyes, enriching for tetracycline-resistant Salmonella, Shigella, or engineered enteric strains. This reduces background growth and swarming, improving the detection and isolation of resistant target colonies.
