Description:
Hektoen Enteric Agar (HE) with kanamycin is a modified selective and differential plate medium in which HE formulation is supplemented with an aminoglycoside to further suppress kanamycin-susceptible background flora while retaining Hektoen's ability to differentiate Salmonella and Shigella from other enteric bacteria. The bile salts and dyes of HE's medium help inhibit most Gram-positives and partially or fully inhibit many non-pathogenic coliforms, favoring the growth of enteric pathogens. HE also contains carbohydrates, and fermenting non-target enterics produce acid shifting colony and medium color to yellow to orange/salmon around the colonies. With carbohydrate fermentation salmonella and shigella act as non-fermenters relying on peptone and as a result, they produce alkaline products that make the colony and surrounding medium green to blue-green. Sodium thiosulfate and ferric ammonium citrate help detect H2S by forming black ferrous sulfide, Many Salmonella will have black centers in their colonies.
Kanamycin is an aminoglycoside that binds the 30S ribosomal subunit, causing misreading of mRNA and inhibiting protein synthesis in susceptible bacteria. The combination of HE and kanamycin suppresses kanamycin-susceptible Gram-negative rods and Gram-positive contaminants that would otherwise survive bile and dyes, enriching for kanamycin-resistant Salmonella, Shigella, or engineered enteric strains.
