Description:
Hektoen Enteric (HE) agar with amphotericin B is a modified, moderately selective medium in which standard HE formulation is supplemented with an antifungal agent to suppress yeast and mold, allowing clearer recovery and differentiation of enteric bacteria from heavily contaminated specimens. Bile salts inhibit most Gram-positive organisms and suppress many non-pathogenic Gram-negative enterics, the combination of bromothymol blue and acid fuchsin reports pH changes. Fermenting non-target enterics produce acid, turning colonies and surrounding medium yellow to orange/salmon. Salmonella and Shigella behave as non-fermenters with carbohydrates under HE conditions, relying mainly on peptone, yielding alkaline reactions that keep colonies and surrounding medium green to blue-green. Sodium thiosulfate and ferric ammonium citrate yield black ferrous sulfide where H2S is produced. Many Salmonella form green/blue-green colonies with black centers.
Amphotericin B is a polyene antifungal that binds ergosterol in fungal cell membranes, forming pores that increase membrane permeability and cause leakage of intracellular contents. Amphotericin B adds a layer of selectivity by inhibiting most yeasts and filamentous fungi that may otherwise grow from fecal, food, or environmental samples and obscure bacterial colonies. Bacterial colony visibility is improved and maintains HE color reactions, particularly in specimens with high fungal loads.
