Description:

Hektoen Enteric Agar (HE) with gentamicin is a modified, moderately selective and differential plate medium used to recover and differentiate Salmonella and Shigella from mixed enteric flora while suppressing gentamicin-susceptible background Gram-negative bacteria in addition to the usual Gram-positive inhibition.  Standard HE  contains carbohydrates that support acid production by non-target enterics such as coliforms. pH dyes such as bromothymol blue and acid fuchsin shift color to show whether the medium has become acidic from carbohydrate fermentation or alkaline from peptone metabolism.  Sugar-fermenting flora acidify the medium and shift the color toward yellow to orange.  Peptone metabolism by Salmonella and Shigella alkalinizes the medium, keeping the colonies and surrounding medium green to blue-green.  Sodium thiosulfate as a sulfur source and ferric ammonium citrate as an indicator yield a black ferrous sulfide precipitate where H2S is produced.

Gentamicin is an aminoglycoside that Inhibits many enterobacterales and other Gram-negative rods, as well as some Gram-positives, by targeting the 30S ribosomal subunit.  When added to HE, it further reduces overgrowth by gentamicin-susceptible commensals and non-target organisms while allowing the growth of gentamicin-resistant Salmonella/Shigella.  By inhibiting gentamicin-susceptible Gram-negative rods and some Gram-positive contaminants, the antibiotic reduces background growth and swarming, improving visualization of target colonies.
