Description:
Hektoen Enteric (HE) Agar with chloramphenicol is a modified, moderately selective and differential plate medium in which standard HE agar is supplemented with a broad-spectrum protein-synthesis inhibitor to suppress chloramphenicol-susceptible background flora while retaining HE’s ability to differentiate Salmonella and Shigella from other enteric bacteria. HE contains bile and salts and dyes that inhibit Gram-positive organisms and partially or fully inhibit many pathogenic coliforms. This allows enteric pathogens that are able to tolerate these conditions to grow. HE also contains carbohydrates, and fermenting non-target enterics produce acid shifting colony and medium color to yellow to orange/salmon around the colonies. With carbohydrate fermentation salmonella and shigella act as non-fermenters relying on peptone and as a result, they produce alkaline products that make the colony and surrounding medium green to blue-green. Sodium thiosulfate and ferric ammonium citrate help detect H2S by forming black ferrous sulfide, Many Salmonella will have black centers in their colonies.
Chloramphenicol is a broad-spectrum bacteriostatic antibiotic that binds the 50S ribosomal subunit and inhibits peptidyl transferase activity, blocking protein synthesis in susceptible bacteria. The combination of HE and chloramphenicol suppresses chloramphenicol -susceptible Gram-negative rods and Gram-positive contaminants that would have survived the biles and salts of HE, enriching for chloramphenicol-resistant salmonella, shigella, or engineered enteric strains. This reduces background growth and improves visualization and isolation of resistant target colonies on the HE differential background.
