The mouse IL-12 ELISA kit is a solid-phase immunoassay specially designed to quantitatively measure mouse IL-12 in serum, plasma, or homogenates. It is based on the Sandwich-ELISA mechanism. IL-12 in the sample is bound to the capture antibody immobilized on the 96-well strip plate and then sandwiched with the biotinylated IL-12 antibody. After the addition of HRP-avidin and TMB substrate, the solution in the wells turns blue. The color reaction is stopped by adding the stop solution into the wells, and the color changes from blue to yellow. The color intensity is positively proportional to the IL-12 bound in the initial step. The IL-12 concentration can be calculated according to the standard curve. This kit is tested with high sensitivity and strong specificity. IL-12 mainly functions to promote the differentiation of Th1 cells and to induce type II interferon (IFN)-γ production. IL-12 is also required for optimal IL-2-dependent proliferation and clonal expansion. It is produced by antigen-presenting cells and plays an important role in host defense against intracellular microbial infection and control of malignancy via its ability to stimulate both innate and adaptive immune effector cells. Significant elevation of IL-12 levels in plasma has been found in many types of atherosclerosis and atherosclerotic cardiovascular disease, including stable angina pectoris (SAP), ST-elevation myocardial infarction (STEMI), and acute myocardial infarction (AMI). IL-12 interacts with its receptor, stimulating JAK2 and TYK2 activatity, activating MAPK and PI3K/Akt signal cascades.
