Description

MacConkey Agar with 50 µg/mL ampicillin sodium salt is a selective and differential medium recommended for the isolation, identification, and enumeration of bacteria. MacConkey agar is one of the earliest media developed for the cultivation of enteric microorganisms from a variety of clinical specimens and is used in the examination of milk, dairy products, and pharmaceutical preparations.

The medium contains peptones that supply nitrogenous and carbonaceous compounds, long-chain amino acids, vitamins, and other essential nutrients. Gram-negative bacteria typically grow well on this medium and are distinguished by their ability to ferment lactose. Lactose fermenting strains appear as red or pink colonies, often surrounded by a zone of acid-precipitated bile. The red color results from acid production during lactose fermentation, which lowers the pH and causes the neutral red dye to change color when the pH drops below 6.8. Non-lactose fermenting strains, such as Shigella and Salmonella, remain colorless and transparent, typically not altering the appearance of the medium.

The addition of ampicillin enhances selectivity by inhibiting the growth of ampicillin-sensitive bacteria, allowing for the isolation of ampicillin-resistant strains. This is particularly useful for isolating bacteria carrying ampicillin resistance genes or for screening recombinant strains in food, dairy, and water samples.
