Columbia Agar Base with Amphotericin B is a nutritionally rich agar medium in which a standard Columbia agar base is supplemented with the polyene antifungal drug amphotericin B to provide strong bacterial growth support while selectively inhibiting most yeasts and molds. Columbia agar base is a complex peptone medium originally devised as a blood-agar base for fastidious organisms such as streptococci, staphylococci, Neisseria, and Haemophilus. The peptones and yeast extract supply amino acids, peptides, nitrogen, vitamins, and other growth factors, supporting luxuriant growth of both fastidious and non-fastidious bacteria. Starch (often corn or potato) serves as a carbon source and can bind toxic metabolites, aiding recovery of delicate organisms, while sodium chloride maintains osmotic balance and agar provides the solid surface for colony development.
Amphotericin B is a polyene antifungal that binds to ergosterol in fungal cell membranes, forming pores that lead to leakage of intracellular contents and cell death. When amphotericin B is added to Columbia Agar Base, the medium becomes selective because most yeasts and filamentous fungi are inhibited, while bacteria (which lack ergosterol) are largely unaffected at those levels. This gives a “bacteria-favoring” selectivity: bacterial colonies can grow robustly on the nutritionally rich Columbia agar base, but fungal contaminants are suppressed or eliminated. Columbia Agar Base with Amphotericin B is used when you want the growth performance and colony morphology of Columbia agar but need to exclude fungal overgrowth in specimens or cultures where fungi are common contaminants.
