Description:

Columbia Agar Base with chloramphenicol is a nutritionally rich, non-blood agar medium in which standard Columbia agar base is supplemented with the broad-spectrum antibiotic chloramphenicol to create a selective environment that suppresses most bacteria while allowing growth of chloramphenicol-resistant bacteria and, in many formulations, favors recovery of fungi.  Columbia agar base is a complex peptone medium originally devised as a blood-agar base to cultivate both fastidious organisms (such as streptococci, staphylococci, Neisseria, and Haemophilus) and non-fastidious bacteria.  The peptones and yeast extract provide amino acids, peptides, nitrogen, B-complex vitamins, and other growth factors that support rapid, luxuriant bacterial growth.  Starch serves as a carbon and energy source and can bind or neutralize toxic metabolites, aiding the recovery of more delicate organisms, while sodium chloride maintains osmotic balance.

Chloramphenicol is a broad-spectrum antibiotic that binds to the 50S ribosomal subunit and inhibits peptidyl transferase activity, blocking peptide bond formation and thereby protein synthesis in susceptible bacteria.  It inhibits many Gram-positive and Gram-negative bacteria but has much less effect on yeasts and molds.  When chloramphenicol is added to Columbia Agar Base, the medium becomes selective: chloramphenicol-susceptible bacteria are suppressed or completely inhibited, while chloramphenicol-resistant bacteria and fungi can still grow.
