Description:
MacConkey Agar with 2.5 µg/mL Amphotericin B is a selective and differential medium recommended for the isolation, identification, and enumeration of bacteria. MacConkey Agar is one of the earliest selective and differential media for the cultivation of enteric microorganisms from a variety of clinical specimens and is used by the Standard Methods for the Examination of Milk and Dairy Products and pharmaceutical preparations.
Peptones in the medium provide nitrogenous and carbonaceous compounds, long-chain amino acids, vitamins, and other essential growth nutrients. Gram-negative bacteria typically grow well on this medium and are distinguished by their ability to ferment lactose. Lactose fermenting strains appear as red or pink colonies, often surrounded by a zone of acid-precipitated bile. The red color results from acid production during lactose fermentation, which lowers the pH and causes the neutral red dye to change color when the pH drops below 6.8. Non-lactose fermenting strains, such as Shigella and Salmonella, remain colorless and transparent, typically not altering the appearance of the medium.
The addition of Amphotericin B, a polyene antifungal agent, suppresses the growth of fungi and yeasts without affecting bacterial colony morphology, allowing for the selective isolation of bacteria from samples that may contain fungal contaminants. This makes the medium especially useful in food, dairy, and water microbiology where fungal contamination is a concern.
