Description
Tryptic Soy Agar (TSA) is a general-purpose, nutrient-rich medium widely used for the cultivation and isolation of a broad spectrum of microorganisms, including both fastidious and non-fastidious species. Its balanced formulation, featuring tryptone and soy peptone, supplies essential amino acids, peptides, vitamins, and minerals, while glucose provides a readily available carbohydrate source. Phosphate buffers the medium, and sodium chloride ensures osmotic balance.
The addition of ampicillin sodium salt at 100 µg/mL transforms TSA into a selective medium, effectively inhibiting the growth of ampicillin-sensitive bacteria and allowing for the isolation and propagation of ampicillin-resistant strains. This makes these plates especially valuable in molecular biology for the selection of recombinant bacteria carrying ampicillin-resistance plasmids, as well as in clinical, food, water, and environmental microbiology for monitoring and studying antibiotic resistance.
TSA plates are also frequently used as a base for enriched or selective media and can be further supplemented with blood or other additives to support the growth of more fastidious organisms.
