Quantification

In this sandwich ELISA, Mouse Soluble Cluster of differentiation 40 ligand,sCD40L in the sample is captured between a pre-coated capture antibody and a detection antibody. Signal intensity is proportional to analyte concentration. Quantification uses a standard curve fitted with Curve Expert software, covering 0.274 ng/ml-200 ng/ml. with a minimum detectable dose of 0.274 ng/ml..

For research use only (RUO). Not intended for diagnostic or therapeutic purposes. Validated in mouse serum, plasma, tissue homogenates, cell culture supernates. matrices only.
Description

The mouse soluble cluster of differentiation 40 ligand (sCD40L) ELISA Kit is used to quantitatively measure mouse sCD40L concentrations in serum, plasma, cell culture supernates, or tissue homogenates. It performs well in important characteristics, including sensitivity and specificity. This assay is based on the sandwich ELISA mechanism and enzyme-substrate chromogenic reaction. The solution color develops proportionally to the amount of sCD40L in the sample. And the intensity of the color can be measured at 450 nm via a microplate reader.

CD40L (CD40LG) mediates a range of activities on B cells, including induction of activation-associated surface antigen, entry into the cell cycle, isotype switching, Ig secretion, and memory generation. CD40–CD40L signaling also plays important roles in monocyte activation and DC maturation. CD40L, especially, has been shown to be a critical and powerful regulator of immune responses in atherosclerosis. In humans, CD40L expression peaks in late-stage atherosclerotic plaques, and its expression is related to features of plaque vulnerability. Many studies have shown that elevated serum levels of the sCD40L are linked to hypercholesterolemia, stroke, diabetes, and acute coronary syndrome (ACS) and predict recurrent CVD.

Alternative Names
Cd40lg; Cd40l; Tnfsf5; CD40 ligand; CD40-L; T-cell antigen Gp39; TNF-related activation protein; TRAP; Tumor necrosis factor ligand superfamily member 5; CD antigen CD154
Function
Cytokine that acts as a ligand to CD40/TNFRSF5. Costimulates T-cell proliferation and cytokine production. Its cross-linking on T-cells generates a costimulatory signal which enhances the production of IL4 and IL10 in conjunction with the TCR/CD3 ligation and CD28 costimulation. Induces the activation of NF-kappa-B. Induces the activation of kinases MAPK8 and PAK2 in T-cells. Mediates B-cell proliferation in the absence of co-stimulus as well as IgE production in the presence of IL4. Involved in immunoglobulin class switching.; Acts as a ligand for integrins, specifically ITGA5:ITGB1 and ITGAV:ITGB3; both integrins and the CD40 receptor are required for activation of CD40-CD40LG signaling, which have cell-type dependent effects, such as B-cell activation, NF-kappa-B signaling and anti-apoptotic signaling.
