Quantification
In this sandwich ELISA, interleukin 1β in the sample is captured between a pre-coated capture antibody and a detection antibody. Signal intensity is proportional to analyte concentration. Quantification uses a standard curve fitted with Curve Expert software, covering 12.5 pg/mL-800 pg/mL with a minimum detectable dose of 6.25 pg/mL.
This kit quantitatively measures mouse IL-1β, a master pro-inflammatory cytokine central to innate immune activation, inflammasome signaling, and fever response. Because IL-1β is often released in sharp, transient bursts following stimuli like LPS challenge or NLRP3 inflammasome activation, accurate capture across both baseline and induced conditions demands a carefully calibrated assay.
Detection Range: The 12.5–800 pg/mL range spans resting-state serum levels in naïve mice through the elevated concentrations typical of acute inflammatory models such as endotoxemia or colitis, minimizing the need for serial dilutions while covering most in vivo and in vitro experimental conditions.

- Food-grade synthetic amorphous silica particles are able to directly initiate the endosomal MyD88-dependent pathogen pattern recognition and signaling pathway in steady-state dendritic cells. The ensuing activation of immature DCs results in de novo induction of pro-IL-1beta. PMID: 28645296
- we assessed the role of RIP3 in synergy with Caspase-1 in the induction of IL-1beta production in BMDM after either LPS/ATP or Chlamydia muridarum stimulation. The possibility of pyroptosis and necroptosis interplays and the role of RIP3 in IL-1beta production during Chlamydia muridarum infection in BMDM was investigated as well. PMID: 28660207
- These data reveal how, upon XIAP deficiency, a TLR-TNF-TNFR2 axis drives cIAP1-TRAF2 degradation to allow TLR or TNFR1 activation of RIPK3-caspase-8 and IL-1beta. This mechanism may explain why XIAP-deficient patients can exhibit symptoms reminiscent of patients with activating inflammasome mutations. PMID: 28723569
