In this sandwich ELISA, Human Interleukin 1α,IL-1α in the sample is captured between a pre-coated capture antibody and a detection antibody. Signal intensity is proportional to analyte concentration. Quantification uses a standard curve fitted with Curve Expert software, covering 3.9 pg/ml-250 pg/ml. with a minimum detectable dose of 0.975 pg/ml..
This human IL1A ELISA kit employs the quantitative sandwich enzyme immunoassay technique to measure the levels of human IL1A in multiple samples, including serum, plasma, cell culture supernates, tissue homogenates, or cell lysates. The enzyme-substrate chromogenic reaction is also used to amplify the signal and quantify the levels of the analyte through the intensity of the colored product. The color intensity positively correlates with the amount of IL1A bound in the initial step.
IL1A is constitutively expressed in epithelial, endothelial, and stromal cells and can be upregulated in hematopoietic and non-hematopoietic cells by diverse stimuli, including TLR agonists, inflammatory cytokines, and oxidative stress. In non-hematopoietic cells, IL1A signals via IL1R on myeloid cells in a MyD88/RIPK1/(TAK1)-dependent manner to induce the expression of pro-inflammatory cytokines. IL1A also plays a role in neutrophil recruitment and host defense after infections caused by viral, fungal, or protozoan. Dysregulation of IL1A production is associated with numerous auto-inflammatory disorders.
