In this sandwich ELISA, interleukin 16 (lymphocyte chemoattractant factor) in the sample is captured between a pre-coated capture antibody and a detection antibody. Signal intensity is proportional to analyte concentration. Quantification uses a standard curve fitted with Curve Expert software, covering 0.156 ng/mL-10 ng/mL with a minimum detectable dose of 0.039 ng/mL.

Description

The ELISA Kit is designed for quantitatively measuring human IL-16 levels in samples, including serum, plasma, cell culture supernates, or tissue homogenates. It uses the sandwich enzyme immunoassay technique in combination with the enzyme-substrate chromogenic reaction to quantify the analyte in the sample. The color develops positively to the amount of IL-16 in samples. The color intensity is measured at 450 nm via a microplate reader.

IL-16 is known to induce the IL-2Rα on T cells, suppress HIV replication and inhibit T-cell antigen receptor/CD3 mediated T-cell stimulation in mixed lymphocyte reactions. In addition, IL-16 acts in concert with IL-2 and/or IL-15 to promote CD4 + T-cell proliferation, and is a chemoattractant factor for CD4 + cells. IL-16 plays a key role in airway hyper-responsiveness and up-regulation of IgE. IL-16 has been associated with diseases characterized by CD4 + T-cell involvement, such as allergic asthma, rheumatoid arthritis, multiple sclerosis, and acquired immunodeficiency syndrome.

Function
Interleukin-16 stimulates a migratory response in CD4+ lymphocytes, monocytes, and eosinophils. Primes CD4+ T-cells for IL-2 and IL-15 responsiveness. Also induces T-lymphocyte expression of interleukin 2 receptor. Ligand for CD4.; may act as a scaffolding protein that anchors ion channels in the membrane.; Isoform 3 is involved in cell cycle progression in T-cells. Appears to be involved in transcriptional regulation of SKP2 and is probably part of a transcriptional repression complex on the core promoter of the SKP2 gene. May act as a scaffold for GABPB1 (the DNA-binding subunit the GABP transcription factor complex) and HDAC3 thus maintaining transcriptional repression and blocking cell cycle progression in resting T-cells.
