What Dulbecco's PBS Is, in the Source's Own Words

Dulbecco's phosphate-buffered saline (DPBS) is a balanced salt solution designed for multiple cell culture applications due to its osmotic balance property similar to the natural condition of mammalian systems. D-PBS is generally used to provide the appropriate milieu to sustain the physiological and structural integrity (i.e., tonicity/viability) of cells in vitro briefly. Balanced Salts support viable conditions for cells only during experimental conditions outside of their regular growth environment. When treated with D-PBS, the cell maintains osmotic balance and physiological pH, and D-PBS protects the cells from acute fluctuations.

A wide range of uses include (a) general cell culture, (b) washing adhesion cells before dissociation, (c) transporting cells or tissue samples, (d) diluting cells for counting, and (e) preparing reagents for providing inorganic salts essential for basic cell metabolism. D-PBS is widely used because its buffering capacity is effective at in vivo neutral pH in various applications. When included with calcium and magnesium, trypsinization / cell dissociation is less effective, but such inclusion is useful for transport media or for reagent preparation. In addition, D-PBS is generally used as a diluent or as an inorganic base for standard media preparation. Deterioration of the D-PBS may be recognized by precipitate or particulate matter throughout the solution, cloudy appearance, color change, and/or pH change.

Scope of the Features list. The water grade, the nuclease and protease statements, and the grade designation above are the Features stated in the source description for the Dulbecco's PBS (D-PBS) line, catalog numbers beginning DCP-. The source lists FNK-87-972 and FNK-87-948 separately, and states no corresponding Features for them; those fields are therefore left blank rather than inferred. Why It Matters

Six Things D-PBS Is Doing While Your Cells Are Off the Incubator

Every point below is drawn from the source description for this category.

water_drop

Osmotic balance close to the mammalian condition

D-PBS is a balanced salt solution designed for multiple cell culture applications because its osmotic balance property is similar to the natural condition of mammalian systems.

science

Buffering at in vivo neutral pH

D-PBS is widely used because its buffering capacity is effective at in vivo neutral pH across a range of applications. Treated cells maintain osmotic balance and physiological pH.

shield

Protection from acute fluctuations

D-PBS protects cells from acute fluctuations while they are held outside their regular growth environment.

timer

A short-term holding solution, by design

Balanced salts support viable conditions for cells only during experimental conditions outside of their regular growth environment. D-PBS sustains physiological and structural integrity — tonicity and viability — briefly.

tune

Calcium and magnesium change what the buffer is for

When included with calcium and magnesium, trypsinization / cell dissociation is less effective — but that inclusion is useful for transport media or for reagent preparation.

visibility

Deterioration is visible before it is measurable

Deterioration of the D-PBS may be recognized by precipitate or particulate matter throughout the solution, cloudy appearance, color change, and/or pH change.

The one decision that changes the result

Both calcium- and magnesium-containing and calcium- and magnesium-free D-PBS are stocked on this page, and they are not interchangeable. The source is explicit: with calcium and magnesium present, trypsinization and cell dissociation are less effective, while that same formulation is the useful one for transport media and reagent preparation. Choose the formulation by the step it has to support, not by what is on the shelf.

Selection Path

Four Questions, in Order, and You Are on the Right Product Page

The catalog above splits along exactly four axes. Answer them in this order — each one narrows the list.

  1. 1

Formulation With or without Ca2+ and Mg2+?

If the next step is washing adhesion cells before dissociation, the source points away from calcium and magnesium: their inclusion makes trypsinization / cell dissociation less effective. If the buffer is going to be a transport medium or a base for reagent preparation, the source calls that inclusion useful.

  2. 2

    Format 1X or 10X?

The catalog lists both a 1X presentation and a 10X presentation for the calcium- and magnesium-containing and the calcium- and magnesium-free formulations. Confirm the working strength and the dilution instruction on the individual product page before ordering.

  3. 3

    Sterility Sterile or Non-Sterile?

On this catalog the 1X items are named Sterile and the 10X items are named Non-Sterile. If the solution will contact a live culture without a further sterilizing step, that naming is the first thing to check.

  4. 4

    Fill Which size?

Every D-PBS configuration is listed in 500 ml, 1000 ml, and 10 liter. The separately supplied Dulbecco's Phosphate-Buffered Saline, PBS(-) is listed in 1000 ml and 12 X 1000 ml. Each size carries its own catalog number, so order against the catalog number, not the product name.

Reading the water specification. The source states the D-PBS line is made with “ASTM 0.05 µS/cm Type 1 Ultrapure H2O | Deionized”. For context, the published ASTM D1193 table sets the Type I ceiling at 0.056 µS/cm maximum electrical conductivity at 298 K, with a minimum electrical resistivity of 18 MΩ·cm; the source figure therefore sits inside the Type I limit rather than contradicting it. Consult the controlling published ASTM table for the full set of Type I constituent limits.
Applications

Where Each Formulation Belongs

Applications below are the ones named in the source description, split by the formulation the source ties them to.

FAQ

Questions Buyers Ask Before They Pick a Configuration

The source lists general cell culture, washing adhesion cells before dissociation, transporting cells or tissue samples, diluting cells for counting, and preparing reagents that provide inorganic salts essential for basic cell metabolism. It is also used as a diluent and as an inorganic base for standard media preparation.

The source states that when calcium and magnesium are included, trypsinization and cell dissociation are less effective, but that the same inclusion is useful for transport media or for reagent preparation. If a dissociation step follows, the calcium- and magnesium-free configuration is the one the source points to.

The source describes D-PBS as sustaining the physiological and structural integrity of cells in vitro briefly, and states that balanced salts support viable conditions only during experimental conditions outside of their regular growth environment. It is not a growth medium. The source states no maximum holding time, so confirm the handling window on the product page.

Both are listed for the calcium- and magnesium-containing and the calcium- and magnesium-free formulations. In this catalog the 1X items are named Sterile and the 10X items are named Non-Sterile. The source gives no dilution instruction and no working-strength figure, so confirm both on the individual product page before ordering.

The source names four signs: precipitate or particulate matter throughout the solution, cloudy appearance, color change, and pH change.

The source states the D-PBS line uses ASTM 0.05 microsiemens per cm Type 1 ultrapure deionized water. For context, the published ASTM D1193 table sets Type I at a maximum electrical conductivity of 0.056 microsiemens per cm at 298 K and a minimum electrical resistivity of 18 megohm-cm, so the figure in the source sits inside the Type I limit rather than contradicting it.

They are a separate supply line and are not FluxMPS items. The source description lists them apart from the DCP catalog numbers and states none of the D-PBS Features for them, so the water grade, the nuclease and protease statements, and the grade designation given on this page do not apply to them. Their formulation attributes are marked as not stated in the catalog above; confirm them on their own product pages.
