Dulbecco's PBS
Description:Dulbecco's phosphate-buffered saline (DPBS) is a balanced salt solution designed for multiple cell culture applications due to its osmotic balance property similar to the natural condition of mammalian systems. D-PBS is generally used to provide the appropriate milieu to sustain the physiological and structural integrity (i.e., tonicity/viability) of cells in vitro briefly. Balanced Salts support viable conditions for cells only during experimental conditions outside of their regular growth environment. When treated with D-PBS, the cell maintains osmotic balance and physiological pH, and D-PBS protects the cells from acute fluctuations.
A wide range of uses include (a) general cell culture, (b) washing adhesion cells before dissociation, (c) transporting cells or tissue samples, (d) diluting cells for counting, and (e) preparing reagents for providing inorganic salts essential for basic cell metabolism. D-PBS is widely used because its buffering capacity is effective at in vivo neutral pH in various applications. When included with calcium and magnesium, trypsinization/ cell dissociation is less effective, but such inclusion is useful for transport media or for reagent preparation. In addition, D-PBS is generally used as a diluent or as an inorganic base for standard media preparation. Deterioration of the D-PBS may be recognized by precipitate or particulate matter throughout the solution, cloudy appearance, color change, and/or pH change.
