Description:
Sabouraud Dextrose Agar (SDA) is an acidic, high dextrose peptone-based medium, used primarily for the isolation, cultivation and maintenance of yeasts and mold. The low pH and elevated glucose content favors fungal growth, while inhibiting bacteria in clinical, cosmetic, environmental and food samples.
How it Works:
The acidic pH of 5.6 is a vital part of SDA's selectivity. The acidic pH directly inhibits the growth of most bacterial species, such as gram positive bacteria which prefer neutral to slightly alkaline conditions. This allows fungi to thrive in the acidic environments. Dextrose provides Fungi with energy and carbon source allowing for favorable growth. Peptones provides nitrogen, amino acids and minerals essential for fungal growth. Fungi are more flexible in nutrient utilization. The medium is usually incubated at around 25-30°C, a range typically optimal for most fungi due to reducing bacterial competion which prefer higher temperatures.
