Description:
Levine EMB (Eosin Methylene Blue) agar is a low selective differential medium used mainly for coliform and enteric Gram-negative isolation and differentiation, especially fecal coliforms such as escherichia coli. The dyes eosin Y and methylene blue interact with the cell walls and membranes of many Gram-positive bacteria reducing their ability to grow or form normal colonies, so Gram-negative rods are favored. Levine EMB contains lactose as the fermentable carbohydrate, bacteria that are able to ferment lactose produce organic acids, lowering the pH of the medium around their colonies. Moderate/slow lactose fermenters produce less acid, often not enough for intense sheen, colonies appear pink to purple or brownish, frequently mucoid with darker centers and paler edges. Non-lactose fermenters do not acidify the medium from lactose, leaving the pH either neutral or slightly alkaline.
