FluxMPS™ Dulbecco's Modified Eagle Medium (DMEM), High Glucose & 50mM HEPES w/o L-Glutamine: 2X Liquid

Double-strength (2X) DMEM High Glucose with 50 mM HEPES — 9.0 g/L glucose and 50 mM HEPES in the concentrate, 4.5 g/L and 25 mM at 1X — for soft-agar, collagen-gel, hydrogel and other 3D or semisolid cultures, ultra-filtered through a quadruple-stage 0.1 µm / 0.04 µm filtration system. With a sodium bicarbonate + 50 mM HEPES dual buffer, it is approximately 5× cleaner than conventional 0.22 µm-filtered media by particulate count.

  • 2X concentrate: mix with an equal volume of a compatible solution or matrix for a final 1X medium
  • Quadruple-stage filtration: 0.1 µm membrane filtered twice, 0.04 µm membrane filtered twice
  • 0.04 µm nano-filtration final polish acts as a mycoplasma barrier
  • 50 mM HEPES (25 mM at 1X) for CO₂-independent pH stability during matrix preparation and handling
  • Endotoxin < 0.05 EU/mL (USP <85> BET)

Why FluxMPS™

Engineered where standard media fails

Conventional DMEM is sterile-filtered once at 0.22 µm. That pore size can pass mycoplasma and leaves subvisible particulates that settle in microchannels, foul membranes and hydrogels, and add noise to optical readouts.[11,12] FluxMPS™ DMEM keeps Dulbecco's formulation and removes that failure mode.

Microchannel-safe purity

A 0.04 µm final filter and USP <788> particulate control keep subvisible debris out of chips, tubing, membranes and matrices.

Purity Architecture

Quadruple-stage filtration system

Every lot of DCP-DMEMH-Q2X is membrane filtered at 0.1 µm twice and 0.04 µm twice — a ready-to-dilute 2X DMEM polished to a 0.04 µm final pore size, a purity level single-pass 0.22 µm media cannot reach.

Applications

Where FluxMPS™ DMEM fits

DMEM High Glucose is a widely used basal medium containing 4.5 g/L glucose at its 1X working concentration.[1,2] It supports many primary and established mammalian cell types, including fibroblasts, neurons, glial cells, endothelial cells, smooth-muscle cells, HEK 293, HeLa, COS-7, and other normal or transformed cell lines.[3] It is commonly used in cell-growth, transfection, recombinant-protein, tissue-engineering, drug-discovery, and cell-signaling research.

DMEM High Glucose 2X is a double-strength formulation containing 9.0 g/L glucose in the concentrate and providing 4.5 g/L glucose after dilution to 1X. It is designed to be mixed with an equal volume of a compatible solution or matrix while maintaining the intended final 1X concentration of glucose, amino acids, vitamins, and salts.

Researchers use the 2X formulation to prepare customized media and cultures containing large volumes of agar, agarose, methylcellulose, collagen, hydrogels, supplements, treatment solutions, or other aqueous components. It is particularly useful for soft-agar colony assays, collagen-gel contraction studies, semisolid cultures, 3D cell culture, stem-cell research, tissue engineering, and other workflows in which adding a matrix or solution would dilute conventional 1X DMEM.[4,5]

Automated Bioreactors & Robotics

Next-Generation System Uptime

For automated bioreactors, liquid-handling robots and closed perfusion loops with fine valves and in-line sensors, an optional 10 nm (0.01 µm) ultra-filtered grade of this medium is available.

  • Total Particulate Exclusion: 0.01 µm filtration removes virtually all particulates down to the nanometer scale.
  • Valve & Sensor Protection: keeps micro-valves, pumps and optical or electrochemical sensors free of deposits.
  • Extended Perfusion Stability: supports long, uninterrupted perfusion runs with fewer line changes.

Note: pH and sodium bicarbonate concentration of the prepared medium are critical factors affecting cell growth. This is also influenced by amount of medium and volume of culture vessel used (surface to volume ratio). For example, in large bottles, such as Roux bottles, pH tends to rise perceptibly as a significant volume of carbon dioxide is released. Therefore, optimal conditions of pH, sodium bicarbonate concentration and surface to volume ratio must be determined for each cell type. We recommend stringent monitoring of pH. If needed, pH can be adjusted by using sterile 1N HCl or 1N NaOH or by bubbling in carbon dioxide. If required, supplements can be added to the medium prior to or after filter sterilization observing sterility precautions.

FAQ

Frequently asked questions

Common questions about using DCP-DMEMH-Q2X for semisolid, 3D, microfluidic and conventional culture.

Yes. DCP-DMEMH-Q2X is quadruple-stage filtered to a 0.04 µm final pore size, so it carries far fewer subvisible particles than 0.22 µm media — approximately 5× cleaner by particulate count. That lowers the risk of clogging microchannels, valves and membranes in organ-on-a-chip, hydrogel-based 3D and microfluidic perfusion systems.[8,9,10]
The medium is filtered four times: 0.1 µm twice and 0.04 µm twice. A single 0.22 µm pass can let mycoplasma and fine particulates through; the 0.04 µm stages act as a mycoplasma barrier and polish out sub-micron particles before ISO Class 5 aseptic fill.[11]
L-glutamine breaks down in liquid medium over time, releasing ammonia that can stress sensitive cultures; a glutamine-free base lets you add fresh L-glutamine or a stable dipeptide just before use. Our complete 2X formulation carries 1,168 mg/L L-glutamine (584 mg/L at 1X). The 50 mM HEPES in the 2X gives 25 mM after 1:1 dilution and adds buffering capacity that does not depend on CO₂, so pH holds steadier while you mix in agar, collagen or hydrogels, handle plates or image on an open stage.[6] The sodium bicarbonate system still works in a CO₂ incubator. Users are advised to review the literature for supplementation and physiological growth requirements specific to each cell type.[1,2]
A CO₂ incubator is recommended once diluted to 1X, because the medium is bicarbonate-buffered (3,700 mg/L at 1X). The 25 mM HEPES at 1X keeps pH steadier during periods in room air.
Mix 2X DMEM with an equal volume of a compatible solution or matrix — agar, agarose, methylcellulose, collagen, hydrogels, supplements or treatment solutions — so glucose (4.5 g/L), amino acids, vitamins and salts end up at their 1X concentration.[4,5] Serum, antibiotics and other supplements can be added before or after filter sterilization, observing sterility precautions.
The endotoxin specification is < 0.05 EU/mL, tested by the USP <85> bacterial endotoxins test (BET) and reported on the lot Certificate of Analysis. The medium is made with Ultrapure Type 1 water (18.2 MΩ·cm).
Yes. Each lot ships with a CoA covering appearance (red), pH 7.4 (± 0.04), sterility (no bacterial or fungal growth after 14 days of incubation per USP), endotoxin and osmolality results. Request copies at support@diagnocine.com.
