Description:
Mueller Hinton Agar (MHA) with 2.5 µg/mL amphotericin B is designed for robust antimicrobial susceptibility testing and the cultivation of most non-fastidious bacterial pathogens. Originally formulated as a clear agar for Neisseria species, MHA is now the standard for determining sulfonamide resistance in gonococci and other organisms, as well as for the diffusion of antimicrobial agents using the Kirby-Bauer disc diffusion method.
The addition of 2.5 µg/mL amphotericin B, a potent antifungal agent, renders the medium selective by inhibiting the growth of fungi and yeasts. This allows for accurate bacterial susceptibility testing and cultivation in samples that may be prone to fungal contamination.
Mueller Hinton Agar is not suitable for disc diffusion assays with slow-growing organisms, anaerobes, or capnophiles, as prolonged incubation can degrade antibiotics and yield imprecise results. It remains the recommended medium for routine antimicrobial diffusion testing using paper discs, now with added antifungal protection for cell culture and clinical microbiology applications.
