Description:

Mannitol Salt Agar (MSA) with tetracycline is a modified MSA in which tetracycline antibiotic is added to provide an extra layer of selection on top of the high-salt and mannitol fermentation system.  MSA contains peptones and beef extract which supply nitrogen, amino acids, vitamins, minerals, and other growth factors required for bacterial growth.  The NaCl inhibits most non-halotolerant bacteria and enriches for salt-tolerant Gram-positive organisms.  D-mannitol is the fermentable carbohydrate and the phenol red acts as a pH indicator.  The medium is red at neutral to alkaline pH and becomes yellow where mannitol fermentation produces acid.

Tetracycline is a broad-spectrum bacteriostatic antibiotic that binds to the 30S ribosomal subunit and blocks attachment of aminoacyl-tRNA, thereby inhibiting protein synthesis in susceptible bacteria.  When tetracycline is added to MSA, it introduces an extra selection layer.  Bacteria that can grow at the high salt concentration and are resistant to tetracycline will grow, so the medium remains selective for salt-tolerant organisms and differential for mannitol fermentation via mannitol and phenol red.
