Description:

Mannitol Salt Agar (MSA) with chloramphenicol is a modified version of standard MSA in which a broad-sprectrum bacteriostatic antibiotic is added to suppress chloramphenicol-susceptible bacteria, while the medium still selects for salt-tolerant organisms and differentiates mannitol fermenters from non-fermenters.  MSA contains peptones and beef extract that supply vitamins, amino acids, minerals, nitrogen, and other growth factors needed for bacterial growth.  NaCl is included and at a high-salt level the medium inhibits most non-halotolerant organisms and enriches for salt-tolerant Gram-positive bacteria such as Staphylococcus spp.  D mannitol is a fermentable carbohydrate and phenol red is a pH indicator with the medium being red at neutral/alkaline pH and turns yellow where mannitol fermentation produces acid.  

Chloramphenicol is a broad-spectrum, primarily bacteriostatic antibiotic that binds to 50S ribosomal subunit and inhibits peptidyl transferase, blocking peptide bond formation and thus protein synthesis in susceptible bacteria.  The antibiotic is acitve against many Gram-positive and Gram-negative organisms.  The combination of MSA and chloramphenicol, it introduces an additional layer of selectivity on top of the already high salt barrier.  Organisms that are able to tolerate the NaCl levels and are resistant to chloramphenicol are able to grow well.
