Description:
Mannitol Salt Agar (MSA) with Carbenicillin is a modified version of standard MSA in which a β-lactam antibiotic (Carbenicillin) is added to further inhibit susceptible background bacteria, while the medium still selects for salt-tolerant staphylococci and differentiates mannitol fermenters from non-fermenters. MSA contains peptones and beef extract which supply nitrogen, vitamins, minerals, amino acids, and other growth factors needed for bacterial growth. The NaCl creates high salt conditions that inhibit most bacteria and allowing salt-tolerant organisms such as Staphylococcus spp to grow. D-mannitol is a fermentable carbohydrate and the phenol red is a pH indicator with the medium becoming red at neutral to alkaline pH and becomes yellow when acid is introduced from mannitol fermentation.
Carbenicillin is a broad spectrum β-lactam antibiotic that targets bacterial cell-wall synthesis by binding to penicillin- binding proteins and inhibiting peptidoglycan cross-linking, that weakens the cell walls and leads to lysis in susceptible cells. Carbenicillin added with MSA further inhibits carbenicillin-susceptible flora that may still be able to tolerate salt.
