Description:

Mannitol salt agar (MSA) with kanamycin is a modified version of standard MSA in which an aminoglycoside antibiotic is added to increase suppression of kanamycin-susceptible background bacteria, while preserving the medium's normal salt selectivity and mannitol-based differentiation for staphylococci.  MSA contains peptones and beef extract, which provides nitrogen, amino acids, vitamins, minerals, and other growth factors needed for bacterial growth.  The NaCl included has a high-salt level that inhibits most non-halotolerant organisms and enriches for salt-tolerant Gram-positive bacteria such as staphylococci and some micrococci.  D-mannitol is the fermentable carbohydrate, and phenol red is the pH indicator.  The medium is red at neutral/alkaline pH and turns yellow around colonies that ferment mannitol and produce acid.

Kanamycin is an aminoglycoside antibiotic that binds to the 30S ribosomal subunit and interferes with translation causing a misreading of mRNA and inhibition of protein synthesis in susceptible bacteria.  It is active against many aerobic Gram-negative rods and some Gram-positive organisms.  The addition of kanamycin to MSA provides an extra layer of selection in addition to the high-salt barrier, so only bacteria that can tolerate the salt concentration and are resistant to kanamycin will grow well.
