This is a recombinant protein in which the Fc region of an antibody is fused to the C-terminus of the human E-cadherin extracellular domain (Asp155 to Lys697), produced in 293 cells. This E-cadherin Fc protein has been purified and orientationally coated on the surface of polystyrene culture plates with the N-terminus of cadherin facing upward. It has been confirmed that multiple types of human iPS cells can proliferate while maintaining their undifferentiated and pluripotent states on the E-cadherin Fc plate.

How To Use This Product:
-
Remove the E-cadherin Fc plate from the bag and add sterilized DPBS (+) to each well. Refer to the below table for the addition volumes.
Cell Culture Area (cm²/well) DPBS (Mg,Ca) Volume (mL/well) 0.35 0.1 -
Prepare the cell suspension for seeding.
-
Remove the DPBS (+) from the E-cadherin Fc plate and immediately seed the cells without letting the plate dry. If the culture surface of the plate dries, the activity of E-cadherin will decrease.
Detachment Method for Human iPS Cells Cultured on This Product
-
Remove the medium from the plate and rinse with sterilized DPBS without calcium and magnesium.
-
Add an appropriate amount of 0.5 mM EDTA in PBS and incubate in a 37°C incubator for about 2-3 minutes.
*Adjust the incubation time depending on the cells used. -
Add medium containing 10 mM Y-27632 to the cells, collect by pipetting, centrifuge, and remove the supernatant.
*Use the dedicated medium suitable for your cells.
