Description

Mueller Hinton Agar (MHA) was originally developed as a simple, clear agar medium for the cultivation of Neisseria species. Over time, its primary use has shifted to determining sulfonamide resistance in gonococci and other organisms. MHA is widely used as a test medium for antimicrobial susceptibility testing (AST), particularly for the diffusion of antimicrobial agents impregnated on paper discs through an agar gel.

The formulation of Mueller Hinton Agar demonstrates excellent batch-to-batch reproducibility for susceptibility testing. It contains low levels of sulfonamide, trimethoprim, and tetracycline inhibitors, supporting the growth of most non-fastidious bacterial pathogens. For antimicrobial susceptibility testing of fastidious organisms like Streptococcus pneumoniae and Haemophilus influenzae, Mueller Hinton Agar supplemented with 5% sheep blood or hemoglobin is recommended.

Mueller Hinton Agar is not suitable for disc diffusion assays involving slow-growing organisms, anaerobes, or capnophiles, as prolonged incubation may degrade antibiotics and yield imprecise results. It remains the recommended medium for antimicrobial diffusion testing using paper discs in routine microbiology laboratories.
