It can simultaneously identify the Omicron and Delta variants by detecting two mutations of spike protein characteristic of the Omicron variant (E484A and N856K) and a mutation characteristic of the Delta variant (L452R).

KANEKA RT-PCR Kit is a reagent intended to detect two types of mutations (E484A and N856K) found in the SARS-CoV-2 Omicron (B.1.1.529) spike protein and the L452R mutation found in the SARS-CoV-2 Delta (B.1.617.1) spike protein by using a one-step RT-PCR (Reverse Transcription Polymerase Chain Reaction) combined with a fluorescent probe. The 3' end of the fluorescent probe contained in the reaction solution has been modified with a quencher and the 5' end with a fluorescent substance (FAM for E484A and N856K detection and ROX for L452R detection). During the process of DNA elongation while acquiring four kinds of deoxyribonucleoside triphosphate, the fluorescent probe is dissolved and the fluorescent substance that has been quenched by the quencher is freed and emits fluorescence. The fluorescence in each PCR cycle is measured with a real-time PCR system to determine whether any gene mutations in the SARS-CoV-2 (Omicron/Delta) spike protein are present or not present based on the Ct level of each fluorescence.
