Description:
Hektoen Enteric (HE) Agar with ampicillin sodium salt is a modified, moderately selective and differential plate medium that combines the standard HE formulation with β-lactam selection to suppress ampicillin-susceptible bacteria while recovering and differentiating ampicillin-resistant enteric targets such as Salmonella or Shigella. HE contains bile salts that are able to inhibit most Gram-positive organisms and many non-pathogenic Gram-negative enterics. Carbohydrates support acid production by non-target fermenters which form yellow to orange/salmon colonies. Peptone utilization by Salmonella and Shigella yields alkaline reactions keeping colonies and surrounding medium green to-blue green, and sodium thiosulfate as well as ferric ammonium citrate detect H2S production. H2S-positive colonies develop black centers or become black.
Ampicillin is a β-lactam that inhibits cell wall synthesis in susceptible bacteria, particularly many Enterobacterales and Gram-positive organisms lacking effective β-lactamases. HE and ampicillin when combined, suppresses ampicillin-susceptible background flora, enriching for ampicillin-resistant Salmonella, Shigella, or other engineered/clinical enteric strains. Functionally, this reduces background growth and potential swarming by susceptible organisms, improving isolation and visualization of resistant target colonies on the HE background.
