Description:
Hektoen Enteric (HE) agar with carbenicillin is a modified moderately selective and differential medium in which HE agar is supplemented with a β-lactam antibiotic to suppress carbenicillin-susceptible bacteria while preserving HE’s ability to differentiate Salmonella and Shigella from other enteric flora. The composition of HE includes bile salts which inhibits most Gram-positive bacteria or fully inhibit many non-pathogenic coliforms and other commensal Gram-negative rods. The carbohydrates and pH indicators distinguish fermenters from non-fermenters. Fermenting non-target enterics produce acid changing the colonies and surrounding medium yellow to orange/salmon. Salmonella and shigella act as non-fermenters and rely on peptone mainly, producing alkaline end products that maintain colonies and the surrounding colony medium green to blue-green. Sodium thiosulfate and ferric ammonium citrate detect H2S by forming black ferrous sulfide and as a result, many Salmonella produce green/blue colonies with black centers.
Carbenicillin is a broad-spectrum β-lactam that inhibits cell wall synthesis, particularly in many Gram-negative rods and some Gram-positives that lack β-lactamases. The combination of HE and carbenicillin suppresses carbenicillin-susceptible background flora (Gram-negative and Gram-positive) while allowing carbenicillin resistant enteric targets to grow. This enhances selectivity beyond bile salts alone, reducing background growth and making resistant Salmonella/Shigella or engineered enterics easier to isolate and read on the HE differential background.
