Description:
Hektoen Enteric (HE) agar with Penicillin G is a modified, selective and differential plate medium in which the standard HE formulation is combined with a β-lactam antibiotic to suppress penicillin-susceptible Gram-positive flora (and some Gram-negatives) while recovering and differentiating enteric bacteria, especially Salmonella and Shigella. HE contains bile salts that inhibit most Gram-positive organisms and suppress many non-pathogenic Gram-negative enterics. Carbohydrates support acid production by fermenting non-target flora, which form yellow to orange/salmon colonies with associated color changes in the surrounding medium. Salmonella and Shigella behave as non-fermenters of these plate carbohydrates under HE conditions, relying mainly on peptone; they produce alkaline reactions that keep colonies and surrounding medium green to blue-green. Sodium thiosulfate and ferric ammonium citrate detect H2S production and many colonies that are H2S positive develop black centers or become black due to ferrous sulfide deposition.
Penicillin G is a β-lactam antibiotic that inhibits cell wall synthesis by binding penicillin-binding proteins, particularly in many Gram-positive organisms and some Gram-negative rods that lack effective β-lactamases. The combination of HE and Penicillin G results in further suppression of penicillin-susceptible Gram-positive flora and some Gram-negative rods, enhancing the selectivity of HE by limiting growth of organisms that might otherwise survive bile salts and interfere with isolation or visualization of resistant target enteric bacteria.
