Description:
Hektoen Enteric (HE) agar with streptomycin sulfate is a modified, moderately selective and differential plate medium in which the standard HE formulation is supplemented with an aminoglycoside antibiotic to further suppress streptomycin-susceptible background bacteria while preserving the characteristic HE reactions that differentiate Salmonella and Shigella from other enteric flora. Bile salts provide a baseline selectivity against most Gram-positive bacteria and some non-pathogenic Gram-negative enterics. Standard HE contains carbohydrates that support acid production by non-target enterics such as coliforms.. pH dyes such as bromothymol blue and acid fuchsin shift color to show whether the medium has become acidic from carbohydrate fermentation or alkaline from peptone metabolism. Sugar-fermenting flora acidify the medium and shift the color toward yellow to orange. Peptone metabolism by Salmonella and Shigella alkalinizes the medium, keeping the colonies and surrounding medium green to blue-green. Sodium thiosulfate as a sulfur source and ferric ammonium citrate as an indicator yield a black ferrous sulfide precipitate where H2S is produced.
Streptomycin is an aminoglycoside that binds the 30S ribosomal subunit, causing misreading of mRNA and inhibiting protein synthesis in susceptible bacteria. When added to HE, streptomycin suppresses streptomycin-susceptible Gram-negative rods and some Gram-positive organisms. This reduces background growth and potential overgrowth or swarming by susceptible organisms, making it easier to detect resistant target colonies on the HE background.
