FluxMPS™ Dulbecco’s Modified Eagle Medium/ Ham's F-12 Nutrient Mixture (DMEM/F-12) (1:1) w/o L-Glutamine, Sodium Bicarbonate: 1X Liquid is a Microfluidics Suitable, quadruple-stage ultra-filtered (0.1 µm ×2 + 0.04 µm ×2) 1X liquid cell culture medium engineered for organ-on-a-chip (OoC), tissue-on-a-chip (ToC), and microphysiological system (MPS) applications. A quadruple-stage train (0.1 µm ×2 + 0.04 µm ×2) reaches a 0.04 µm final cut-off, five times finer than the 0.22 µm membranes used for conventional sterile filtration. L-Glutamine and sodium bicarbonate are omitted so researchers can supplement precisely to their protocol.

  • Quadruple-stage filtration: 0.1 µm ×2 (Prefiltration I & II) + 0.04 µm ×2 (Final filtration I & II — Polish), reaching a 0.04 µm final cut-off
  • Endotoxin release specification: < 0.05 EU/mL (LAL assay, USP <85> BET), tested per manufacturing batch
  • DMEM/F-12 (1:1) base with 3151 mg/L D-glucose and 110 mg/L sodium pyruvate; pH 7.4 (USP <791>)
  • Formulated without L-glutamine and without sodium bicarbonate — supplement L-glutamine and a buffering system (HEPES or bicarbonate) to match your culture system
  • Contains phenol red (8.630 mg/L) as a visual pH indicator

Why FluxMPS™

Engineered where standard media fails

Conventional 0.22 µm-filtered media carry mycoplasma-sized particulates (0.2–0.3 µm), subvisible debris, and endotoxin variability that accumulate inside microchannels — corrupting biosensor readings, triggering inflammation in primary cultures, and shortening device lifetimes. FluxMPS™ addresses these failure modes through validated four-stage filtration.

filter_alt

Microchannel-safe purity

0.04 µm final filter retains particulates to sub-mycoplasma size; USP <788> Method 1 particulate compliance verified per batch.

Purity Architecture

Quadruple-stage filtration system

FluxMPS™ Dulbecco’s Modified Eagle Medium/ Ham's F-12 Nutrient Mixture (DMEM/F-12) (1:1) w/o L-Glutamine, Sodium Bicarbonate: 1X Liquid is processed through a four-stage serial filtration sequence reaching 0.04 µm — addressing mycoplasma-sized particulates, subvisible particles, and bioburden that 0.22 µm filtration alone does not retain.

  1. 1

    0.1 µm Prefiltration I

Removes large particulates and aggregates; protects the first 0.04 µm final filter cartridge and extends filter life.

  2. 2

    0.04 µm Final filtration I

Fine particulate and bacteria retention; the pore size is smaller than the 0.2–0.3 µm mycoplasma diameter, a class of particulate not addressed by standard 0.22 µm filtration.

  3. 3

    0.1 µm Prefiltration II

Second dedicated prefilter, protecting the second 0.04 µm final filter cartridge in this paired train.

  4. 4

0.04 µm Final filtration II — Polish

Ultimate polishing filter; ISO Class 5 aseptic fill in a validated laminar-flow workstation.

Performance vs. conventional media

Four sequential stages reaching 0.04 µm deliver approximately 5× cleaner media by particulate count compared to single-pass 0.22 µm filtration, with USP <788> Method 1 compliance verified on every batch.

Sterility assurance: Every batch undergoes 14-day USP <71> sterility testing. Mycoplasma control is achieved through 0.1 µm mycoplasma-retentive filtration; this is a filtration statement, not a per-lot mycoplasma assay.
Grade: This product is Microfluidics Suitable, filtered to a 0.04 µm final cut-off. It is not an MPS Grade product — that designation is reserved for the 0.01 µm ultra nano-filtered line, which adds 0.02 µm and 0.01 µm stages after the 0.04 µm polish. Micro-Batch Precision

Small-batch manufacturing ensures lot-to-lot nutrient consistency for reproducible perfusion studies.

Batch-level quality control. Endotoxin is controlled per manufacturing batch rather than per unit. Every batch is tested before release and must meet the release specification:
  • Endotoxin — LAL assay, USP <85> Bacterial Endotoxins Test; assay sensitivity 0.005 EU/mL; release specification < 0.05 EU/mL
  • pH, osmolality, conductivity, appearance and clarity
  • Sterility
A Certificate of Analysis is available on request.

Endotoxin — USP <85> BET

LAL assay per batch. Release specification: < 0.05 EU/mL.

Particulate — USP <788> Method 1

Light obscuration particle count confirms ≥10 µm and ≥25 µm compliance per batch.

Osmolality — USP <785>

Freezing-point osmometry per USP <785>. Result: See CoA.

Documentation — CoA & Full Lot Records

Certificate of Analysis with full QC panel, traceability, and release signatures for every batch.

FAQ

Frequently asked questions

Common questions about FluxMPS™ Dulbecco’s Modified Eagle Medium/ Ham's F-12 Nutrient Mixture (DMEM/F-12) (1:1) w/o L-Glutamine, Sodium Bicarbonate: 1X Liquid and Microfluidics Suitable cell culture media.

Yes. DCP-DM12-QBT1X uses Diagnocine's Quadruple-stage filtration (0.1 µm ×2 + 0.04 µm ×2), delivering ultra-low particulate counts that reduce microchannel clogging risk in OoC and MPS devices.
Standard 0.22 µm filtration does not retain mycoplasma-sized particulates (0.2–0.3 µm). FluxMPS™ uses four sequential stages reaching 0.04 µm, delivering approximately 5× lower particulate counts per USP <788> Method 1.
L-Glutamine is unstable in liquid storage and is omitted so you can add it fresh (typically 2–4 mM, or a stabilized dipeptide alternative) immediately before use. Sodium bicarbonate is omitted to give you control over the buffering system; add sodium bicarbonate for standard 5–10% CO₂ incubation, or HEPES (10–25 mM) for non-CO₂ or open-air culture. Contact support@diagnocine.com for a custom pre-supplemented formulation.
This formulation does not contain sodium bicarbonate or HEPES. CO₂ incubation is therefore optional and depends on the buffering system you add: supplement with sodium bicarbonate for standard CO₂ incubation, or with HEPES (10–25 mM) for non-CO₂ culture. Validate empirically for your cell line.
Yes. This medium can be supplemented with FBS (5–20%), growth factors, antibiotics, or other additives per standard practice. When adding serum or other protein-containing supplements, filter with a 0.2 µm low-protein-binding PES or PVDF membrane; do not use a 0.04 µm membrane, which will strip serum of its functional protein and lipoprotein fraction. Add supplements immediately before use.
Endotoxin is controlled per manufacturing batch by LAL assay per USP <85> BET (assay sensitivity 0.005 EU/mL). Release specification: < 0.05 EU/mL. Every batch is tested before release and results are reported on the Certificate of Analysis, available from support@diagnocine.com.
Yes. A batch-specific CoA is available for every shipment and includes: appearance, pH (USP <791>), osmolality (USP <785>), endotoxin (USP <85> BET), sterility (USP <71>), particulate matter (USP <788> Method 1), raw material traceability, manufacturing date, and authorized release signatures.
