Salmonella Shigella (SS) agar is a moderately selective and differential culture medium used in clinical, food diagnostics, and sanitary bacteriology for the isolation and presumptive identification of pathogenic enteric bacteria such as Salmonella and certain strains of Shigella species.
How it Works:
Salmonella Shigella (SS) agar achieves its selective and differential properties through carefully orchestrated combinations of chemical components that exploit the physiological differences beteween target pathogens and competing microorganisms. The medium functions as a mult layered selection system operating through both exclusionary mechanisms and identification mechanisms. The selectivity of SS agar depends on three complementary inhibitory compounds that work well together to supress gram-positive bacteria, most coliforms, and swarming proteus strains. At the same time this allows Salmonella to grow uninhibited, while some shigella strains may be inhibited by the brilliant green component.
Bile salts acts as the primary selective agent and disrupts the integrity and structure of bacterial cell membranes. Brilliant Green Dye potentiates the inhibitory effects of bile salts. Sodium Citrate functions as a tertiary selective agent by chelating divalent cations (Ca²? and Mg²?) and creating an ionic environment hostile to coliforms and swarming proteus species.
