Description

OptiVitro®CHO CA01β Feed Media SF is a dry powdered cell culture feed supplement, it’s fit for fed-batch processes of Chinese hamster ovary (CHO) cell lines to assist process development and improve the utilization rate of bioreactor. The product is chemically defined (CD), animal-derived component-free (ADCF), and without any growth factors (such as insulin), peptides, hydrolysates, phenol red, 2-mercaptoethanol or other components undefined. It should be used with OptiVitro®CHO CA01α Feed Media SF (Catalog no.:CA000-N011) together, which can increase the yield of monoclonal antibodies and other proteins in CHO fed-batch culture process. This product is with poloxamer 188 and  without L-glutamine.

Prepare media

1. 1.Take the preparation of 1L liquid medium as an example. Measure 80% of the final volume WFI in a clean vessel.
2. 2.Add 108.76g CHO CA01β Feed Media SF powder slowly to the vessel, mix for 60 minutes.
3. 3.Slowly add 115 mL of a 10N NaOH solution（or 46g solid NaOH）, adjust the pH to between 10.9 and 11.3 and mix for 60 minutes.
4. 4.Adjust the pH to between 10.8 and 11.4, stirring 10 minutes.
5. 5.Adjust the final volume to 1L, continue stirring for an additional 10 minutes.
6. 6.Measure and record the final osmolality. Expected values: 300 to 370 mOsmol/kg (diluted 1:5).
7. 7.Sterilize immediately by 0.22μm membrane filtration. Stored for 3 mouths at 2°C to 8°C away from light.

 

• Note:It is necessary to add  NaOH solid many times slowly , so as to avoid excessive exothermic influence on the performance of the medium when NaOH is dissolved.The solution will remain cloudy before adding NaOH solution to adjusting pH,which was a normal phenomenon. It should be clear when NaOH solution is added .
