Methyl Green Solution
Cat No.: DCP-MG0.1X
Size: 500 mL
Sterile, Ultrapure, Filtered 0.1-micron membrane TWICE and 0.04-micron membrane ONCE.
Description:Methyl Green 0.1% w/v aqueous solution is a cationic triphenylmethane dye widely employed in biological staining and molecular biology applications. This ready-to-use formulation is optimized for nuclear counterstaining, DNA visualization, and compatibility with advanced detection systems. With a clear blue-green appearance and pH-dependent color transition (yellow at pH 0.1 to blue at pH 2.3), it serves as a critical tool for researchers requiring precise nucleic acid differentiation and cellular imaging.
Key Features:
- Ready-to-use nuclear counterstain optimized for histology, cytology, and molecular biology applications.
- Pre-purified via chloroform extraction to remove crystal violet contaminants, ensuring consistent staining quality.
- Exhibits far-red fluorescence when bound to DNA (excitation: 633 nm; emission: 677 nm), enabling high-resolution confocal imaging.
- Non-intercalating DNA binding via electrostatic interactions with AT-rich regions, preserving nucleic acid integrity.
- Compatible with aqueous and non-aqueous mounting media, including resin-based formulations.
Applications:
1. Nuclear Staining in Histology/Cytology:- Provides crisp light-green nuclear contrast in formalin-fixed, paraffin-embedded (FFPE) tissues and frozen sections.
- Pairs with chromogenic substrates (e.g., DAB) in immunohistochemistry (IHC) and in situ hybridization (ISH).
- Used in Methyl Green-Pyronin staining to distinguish DNA (green) from RNA (red) in cellular samples.
- Serves as a photostable, far-red fluorescent marker for live-cell nuclei imaging and thick tissue sections.
- Functions as a tracking dye in acidic buffer systems for DNA gel electrophoresis.
- Ideal for teaching laboratories and clinical settings requiring reliable nuclear visualization.
