Description:
Mannitol Salt Agar (MSA) with ampicillin sodium salt is a modified selective and differential medium where ampicillin is added to standard MSA to further suppress ampicillin-susceptible background flora while still recovering salt-tolerant staphylococci. It is not a standard commercial formulation, so any use (including concentration) should be validated in-house for your strains and application.
MSA contains peptones and beef extract which is a source of vitamins, minerals, amino acids, and nitrogen. Sodium chloride produces high osmotic pressure that most bacteria are unable to tolerate. D-mannitol is a fermentable carbohydrate, phenol red is used as a pH indicator, yellow at acidic pH, red at neutral to alkaline pH. Ampicillin Sodium Salt is a β-lactam antibiotic with activity against a broad range of Gram-positive and Gram-negative bacteria. The addition of ampicillin sodium salt to MSA provides additional selective pressure by inhibiting ampicillin-susceptible organisms that might otherwise grow on the high-salt medium alone.
